Chlorophyll fluorescence imaging allows us to visualize how efficiently absorbed light energy is being utilized by the photosynthetic apparatus, particularly within Photosystem II (PSII), providing a sensitive indicator of photosynthetic performance.
Importantly, this technique is non-destructive, non-invasive, and highly sensitive, enabling the early detection of plant stress and physiological changes long before visible symptoms appear.
As a result, chlorophyll fluorescence imaging has become a widely adopted tool in plant physiology, stress biology, crop improvement, and high-throughput phenotyping research, supporting both fundamental studies and agricultural applications.
The FluorCam system is based on Pulse-Amplitude-Modulated (PAM) chlorophyll fluorescence measurement, a widely established technique for assessing photosynthetic function.
During measurement, the system exposes the plant to a controlled sequence of measuring light, actinic light, and saturating light pulses. By analyzing fluorescence responses under these defined light conditions, it quantifies the flow and dissipation of absorbed light energy within the photosynthetic apparatus.
This approach enables the calculation of key photosynthetic parameters, which provide quantitative indicators of photosynthetic efficiency, photoprotective activity, and plant physiological status.
Because PAM fluorescence measurements are non-destructive, non-invasive, and highly reproducible, they provide reliable physiological information without damaging the sample.
